Biosynthesis of Glycoproteins

چکیده
The presence of carbohydrate in many plasma and tissue proteins is well established (1, 2). The glycoproteins thus far studied contain one or several oligosaccharide units linked to a peptide chain by uncharacterized linkages. Little is yet known about the biosynthetic processes by which the oligosaccharide units are assembled and attached to the polypeptide chain. In order to approach these problems, we have investigated the incorporation of glucosamine-l-l% into the protein of cytoplasmic fractions of liver and into plasma protein of the intact rat hoping to obtain information on the subcellular site of incorporation. The incorporation of glucosamine into macromolecules of other tissues has also been studied.

L-fucose (6.deoxy. L.galactose) is the only monosacharide which is placed in non reducting position of glycoproteins structure in L-deoxy form. L-fucosidase and fucosy 1 transferase directly take part in the elevation or decrease of sorum L-fucose levels. Tumors may directly induce enzymes that release L-fucose from glycoproteins formed in the neoplastic tissue, or the glycoproteins may be rele...

Introduction: The development of an effective vaccine against Hepatitis C virus (HCV) is still a target of intensive vaccine research. The HCV envelope proteins E1 and E2 which can induce broadly neutralizing antibodies are the major candidate for this purpose. Different types of expression systems have been used to express these glycoproteins. In this study, an expression system using Pichia p...

Metastasis describes the ability of a tumor to invade normal tissue and generate secondary tumors at sites distant from the primary tumor. The mechanism responsible for the inhibition of tumor metastasis by different agents is at least partly associated with the ability to interfere with cellular functions such as adhesiveness, motility and invasiveness. Certainly effective treatment of metasta...

متن کامل2,6-branched mannose and the regulation of poly-N-acetyllactosamine biosynthesis in N-linked oligosaccharides of Chinese hamster ovary cells.
Poly-N-acetyllactosamine (PL) sequences (repeating (3Gal beta 1-4GlcNAc beta 1)n) in complex-type N-linked oligosaccharides often occur in branched tri- and tetraantennary chains containing alpha-linked mannosyl residues disubstituted by N-acetyllactosaminyl units at C-2 and C-6 (2,6-branched mannose). We report here our studies on the factors affecting PL biosynthesis and the branching of N-li...
متن کاملCerulenin blocks fatty acid acylation of glycoproteins and inhibits vesicular stomatitis and Sindbis virus particle formation.
Cerulenin, an antibiotic that inhibits de novo fatty acid and cholesterol biosynthesis, effectively inhibited the formation and release of virus particles from chicken embryo fibroblasts infected with Sindbis or vesicular stomatitis virus (VSV). When added for 1 h at 3 h postinfection, the antibiotic blocked VSV particle production by 80 to 90% and inhibited incorporation of [3H]palmitic acid i...
